A série, sobre jovens estudantes de escolas de elite (Constance Billard para garotas e St. Jude’s para rapazes) no bairro Upper East Side de Manhattan em Nova Iorque, segue Serena van der Woodsen (Blake Lively) a partir do seu retorno à cidade após fugir e ter se mantido fora do alcance de amigos e conhecidos. Os acontecimentos e histórias de cada personagem são narrados por uma anônima (voz por Kristen Bell), que atendendo pelo pseudônimo Gossip Girl (Garota Fofoca em tradução livre) é autora do blog de mesmo nome onde registra os mesmos. Surpresa com a volta da melhor amiga, Blair Waldorf (Leighton Meester) se contenta excluindo socialmente e mantendo distância de Serena depois da perda de contacto durante sua ausência. O círculo jovem é completado pelo namorado de Blair, Nate Archibald (Chace Crawford) e seu amigo de infância, Chuck Bass (Ed Westwick), e os irmãos até então desconhecidos Dan (Penn Badgley), Jenny Humphrey (Taylor Momsen) e Vanessa Abrams (Jessica Szohr). Fazendo parte da trama há o irmão de Serena, Eric (Connor Paolo), internado em uma clínica de reabilitação, e sua mãe Lily van der Woodsen (Kelly Rutherford), como também Rufus Humphrey (Matthew Settle), separado de sua esposa Alison (Susan Misner), pai e mãe de Dan e Jenny.
